Longterm adverse development effects of methylphenidate treatment for ADHD

EMA’S PROPOSED OBJECTIVES:
 
1) Study of long-term developmental effects including growth
 
2) Study of long-term effects on sexual development and fertility
 
3) Design of appropriate tools to study developmental effects
 
 
 
PRIMARY OBJECTIVES OF ADDUCE WP 5:
 
1) To determine whether methylphenidate treatment for ADHD is associated with a statistically significant increase in long-term (> 1 year) risk of negative effects on:
 
    A) The rate of growth (i.e. height and weight)
 
    B) The rate of pubertal maturation
 
2) To explore the application of intensive monitoring of bone age as a tool to study adverse developmental effects of methylphenidate
 
 
 
SECONDARY OBJECTIVES OF ADDUCE WP 5:
 
1) To explore the role of potential mediators, moderators and confounders (age, sex, socioeconomic status, chronic diseases, family functioning, perinatal conditions and parental height) in the relationship between methylphenidate treatment and adverse outcomes on growth and pubertal maturation
 
2) To describe the dose-response relationship (dosage, duration of treatment, discontinuation vs. continued use) between methylphenidate exposure and risk of adverse effects on growth and pubertal maturation
 
3) To estimate through consensus conference the clinical significance of adverse effects on growth and pubertal maturation in terms of their impact on prescribing practice and adherence/concordance
 
4) To develop a consensus evidence-based clinical guideline to aid the prevention, early identification and management of any adverse effects on growth and pubertal maturation associated with methylphenidate treatment for ADHD
